According to Reborn, Yamamoto has a natural talent for fighting. He claims that Yamamoto is a natural born hitman. Which in many senses may be true. Yamamoto is the only of Tsuna's guardians (aside from Tsuna himself) who didn't have prior training in any form of fighting. Despite this Yamamoto is one of the only guardians who defeats a Varia member during the ring battles and he is considered the second strongest... I believe... when Fuuta ranks the best fighters at Namimori Jr. High for Mukuro.
Yamamoto quickly picks up swordsmanship and claims that it's just like baseball. He is the baseball team's batter and actually used a baseball bat as a weapon prior to having Reborn give him a transforming sword. Yamamoto wasn't ever professionally trained in swordsmanship until a week or so before the Varia Arc.
Even considering that Yamamoto is able not only to stand on par with Squalo, a swordsman who can defeat almost everyone who he comes in contact with just by using the element of surprise and his confidence in his own abilities. Yamamoto can even stop his blade prior to striking a death blow which is a skill that is far beyond that of an amateur swordsman. All in all even though it may not have been his ideal choice of a profession, being a hitman is something he can and would succeed at.
